EM Unit Physiology

Functionally diverse metabotropic GABAB and glutamate receptors (mGluRs) control neuronal excitability and synaptic transmission by activating or inhibiting various types of ion channels, such as high voltage-activated Ca2+, as well as K+ channels. The impact of receptor activation on synaptic integration and regulation of transmitter release depends on the spatial relationship and coupling of receptors and their effectors in subcellular compartments of the target neurons. We have been investigating the structural and functional basis of metabotropic receptor-effector ion channel complex-mediated signaling in cortical principal cells and GABAergic interneurons, as well as studying the activity-dependent regulation of the surface dynamics of protein complexes using a combination of pharmacological and high-resolution quantitative immunoelectron microscopic approaches.

Applications

  • Freezing of large tissue specimens without requiring structure-altering freeze protective additives.
  • Ideally suited for subsequent replication by freeze fracturing in a freeze etching system for TEM applications or subsequent cryo sectioning for cryo SEM and TEM investigations.
  • Best suited in conjuction with subsequent freeze substitution, followed by low temperature embedding and polymerisation for sectioning in a conventional ultramicrotom.
  • Suitable for samples destined for subsequent thin sectioning in their frozen state by cryo-ultramicrotom for cryo-TEM analyses.

  • Freeze-fracturing of cell suspensions and tissues frozen on copper carriers by cryo-jet or plunge-freezing, using the specimen table for copper carrier plates.
  • Freeze-drying of cell components or isolated membranes and viruses and DARS [4].
